DNA replication in eukaryotic cells is dependent on the phosphorylation of the pre-replicative complex (preRC) at the origin of replication. Two complexes of proteins mediate this event, the cyclin dependent kinase (CDK) complex, and the Cdc7 kinase-ASK complex. The activity of Cdc7 kinase oscillates during cell cycle. The major targets of Cdc7 kinase are proteins that belong to the MCM complex (mini chromosome maintenance proteins). Cdc7 kinase was also found to be important in meiosis, checkpoint responses, maintenance of chromosome structure, and repair. Function: Seems to phosphorylate critical substrates that regulatethe G1/S phase transition and/or DNA replication. Canphosphorylates MCM2 and MCM3. Subunit: Forms a complex with either DBF4/DBF4A or DBF4B, leadingto the activation of the kinase activity. Subcellular Location: Nucleus. Similarity: elongs to the protein kinase superfamily. Ser/Thrprotein kinase family. CDC7 subfamily. Contains 1 protein kinase domain. Antigen retrieval: citrate buffer ( 0.01M, pH 6.0 ), Boiling bathing for 15min; Block endogenous peroxidase by 3% Hydrogen peroxide for 30min; Blocking buffer (normal goat serum,C-0005) at 37℃ for 20 min;
Incubation: Anti-Cdc7 Polyclonal Antibody, Unconjugated(bs-5148R) 1:200, overnight at 4°C, followed by conjugation to the secondary antibody(SP-0023) and DAB(C-0010) staining
Blank control(blue): 293T(fixed with 2% paraformaldehyde (10 min)). Primary Antibody: Rabbit Anti-Cdc7/FITC Conjugated antibody (bs-5148R /FITC), Dilution: 1μg in 100 μL 1X PBS containing 0.5% BSA; Isotype Control Antibody: Rabbit IgG/FITC(orange) ,used under the same conditions.
